SRX22799028: whole-genome sequencing of the Corynebacterium amycolatum strain ICIS 82 isolated from the vaginal contents of healthy women

Design: The DNA library for whole-genome sequencing was prepared using a NEBNext Ultra II FS DNA Library Prep Kit for Illumina (New England BioLabs, USA). The library was validated using capillary electrophoresis on a QIAxcel Advanced System (Qiagen, Germany) using the QIAxcel DNA High Resolution Kit and normalized using qPCR on the CFX Connect Real-Time PCR System (Bio Rad, USA). Paired-end sequencing (2250 bp) was carried out on a MiSeq platform (Illumina, USA) using Reagent Kit v.2 (Illumina, USA) in the Center of Shared Scientific Equipment Persistence of microorganisms at the Institute for Cellular and Intracellular Symbiosis UrB RAS.

The genus Corynebacterium spp., also known as "diphtheroids" or "coryneform" includes more than 88 species. Of these, 53 species are occasional or extremely rare causes of diphtheria and diphtheria-like diseases in humans or are transmitted to humans by zoonotic contact. Along with the fact that microorganisms of the genus Corynebacterium play a role in the development of infection in humans, recently, the information about the important role of individual representatives of this group of microorganisms in the formation of microbiocenosis of different biotopes of the human body. Is no exception and the reproductive tract of women, where corynebacteria, according to some researchers, along with lactobacilli play an important role in maintaining colonization resistance. Along with the fact that non-pathogenic corynebacteria have their own antagonistic activity against pathogenic and conditionally pathogenic microorganisms, they potentiate the antagonistic activity of peroxide producing lactobacilli that probably is one of the key mechanisms for the protection of the reproductive tract of women from the development of infections. This project includes the non-pathogenic strains Corynebacterium spp. isolated from the reproductive tract of healthy women which has a good probiotic properties, due to the production of surfactants (biosurfactants) and increased antagonist activity of peroxide producing lactobacilli in relation to opportunistic pathogens.
